Apr, 2007 - Apr, 2007酸素および親電子性分子と総称される一群の毒性化学物質は空気・食物などから体内に取り込まれ、癌、動脈硬化、糖尿病などの原因となる。一方、動物細胞は酸素毒性や親電子性分子を感知する仕組み備えており、細胞内に活性酸素や親電子性分子が出現すると、抗酸化応答系や解毒代謝酵素群が働き、これらのストレスは速やかに消去される。このような適応・応答機構の存在は以前から知られていた。しかし、細胞がどのように酸化ストレスや親電子性分子を感知し、それらに対抗する生体防御遺伝子群の誘導・活性化をしているかは長年の謎であった。代表者らは、Keap1が分子センサーとして親電子性分子や酸化ストレスを感知し、転写因子Nrf2を活性化することで生体防御酵素群の発現を制御していることを発見した。本研究では、生体防御センシングの中核をなすKeap1-Nrf2による酸化ストレス感知とNrf2-MafによるDNA配列認識と転写活性化の分子機構を構造生物学(X線、電子線単粒子解析)と細胞生物学の手法を駆使して解明する。
