Molecular systematics and evolution of the ectomycorrhizal basidiomycete Trichololma matsutake and its allied species on the basis of retrotransposons, including the first report of LTR (marY1) and non-LTR retrotransposons (maryY2N), and an Alu-like short interspersed nuclear element dimer (megB1) in the phylum Basidomycota. The LTR was expressed in Saccharomyces coercive, and the LTR-based vector was amplified in Lentinula edodes after transformation, indicating marY1 is functional. On the basis of the genomic localizations of megB1, it was hypothesized that species belonging to Basidomycota could have evolved in two pathways: one having megB1 in rDNA IGS1 as a major component and the other having megB1 outside rDNA, the localization profiles that do not correlated to current systematics in the phylum. By using retrotransposons as genetic markers, a PCR-based mathod was developed to differentiate among biogeographical populations of Tricholoma matsutake in the Far East.

The occurrence of the genetic mosaicism in the rhizospheric colonies “shiro” through spore dispersal of Tricholoma matsutake has been proven, which otherwise appears to be persistent by vegetative growth of clonal hyphae. Similarly, the occurrence of the genetic mosaicism in cultivated mushrooms using Pholiota nameko was proven wherein genetically heterologous dikayotic mycelia co-ordinate to each other to produce fruiting bodies, a phenomenon that often confers deterioration of commercial spawns when the co-ordination tend to a negative trait. Genetic mosaicism of mushroom producing fungi somewhat resembles biofilm formation in prokaryotes in which autoinducers may work in such a phenomenon.

The Pincceae-specific ectomycorrhizal symbiont Tricholoma matsutake was experimentally proven to associate with Betula platiphylla in vitro as an ectomycorrhizal symbiont, suggesting the fungus could associate with both pine and birch in mixed forests productive of matsutake mushrooms in wild. Also, Tricholoma matsutake was proven to associate with the arbuscular-mycorrhizal plants in Meliaceae and Rosaceae carrying ectoderm in vitro as a root endophyte symbiont, suggesting the fungus innately can associate with a wide range of plants if environmental condition is suitable, and switching could occur between root endophyte association and ectomycorrhiza symbiosis depending upon host plants. Sullies luteus also established root endophyte symbiosis with Prunus speciosa.

Microscopic and genetic analyses showed that Tricholoma matsutake generates basidiospores of homo-dikayon, hetero-dikaryon and monokaryon. Most of monokaryotic hyphae of Tricholoma matsutake become dikaryon without mating in the course of vegetative growth.

Working with RIKEN Ion Beam Breeding Team, Nishina Center for Accelerator-based Science, we succeeded in isolating a pleiotropic mutant of Tricholoma matsutake using argon-ion beam irradiation; the mutant not only exhibited a different colony morphology to the wild-type, but also showed increased degradation of dye-linked water-insoluble amylose and cellulose substrates. With gamma-ray irradiation, a mutant of T. matsutake has been generated whose plant-associating behavior has been converted from beneficial to harmful to its symbiotic partner Pinus densiflora, the finding useful in elucidating mechanisms involved in ectomycorrhizal symbiosis.
