The accumulation of particular protein aggregates is characteristic of age-related neurodegenerative diseases. Recently, biochemistry approaches identified the dominant proteins constituting the protein aggregates. On the other hand, genetical approach identified the gene mutations of the proteins as a cause of the familial neurodegenerative diseases. TDP-43 is a nuclear protein identified as the dominant protein constituting the protein aggregates which accumulate in the cytoplasm of neuronal or glial cells in the brain of nearly half of patients of frontotemporal lateral degeneration (FTLD) or most of patients of amyotrophic lateral sclerosis (ALS). Neuropathological studies showed that the accumulation of TDP-43 is highly correlated with the neurodegeneration. Furthermore, cell biology studies showed that physiological role in nuclear TDP-43 is essential for maintaining cellular function. These findings indicate that the mechanism of TDP-43 accumulation is the key to develop the cure of FTLD/ALS. Therefore, our Lab focus on how or when TDP-43 accmulation starts.
