EFFECTS OF A BRIEF TREATMENT OF STROKE-PRONE SHR WITH CANDESARTAN AT PREHYPERTENSIVE STAGE ON HYPERTENSIVE END-ORGAN DAMAGE IN THE ADULTS AND LIFE-SPAN OF THEIR OFFSPRING

To investigate the effects of prehypertensive short-term treatment with candesartan, we administered candesartan (1mg/kg/day) to male SHRSP from 5 to 10 weeks of age. Pathophysiological examination was carried out at 27 weeks of age. Furthermor, candesartan was administered to both male and female SHRSP at the same manner and then brother-sister breeding was performed at 11 weeks of age and the lifespan of the offspring was monitored. Hypertensive end-organ damage was markedly suppressed in the treated group. PRA, plasma A II levels, and cerebral ACE activity were significantly lower in the treated group. Lifespan in the offspring obtained from treated group was significantly longer as compared to that in the control group. These results demonstrated that early treatment with candesartan for 5 weeks is effective not only for the prevention of hypertensive end-organ damage after maturation but also prolonged the lifespan of their offspring.
