© 2014 Elsevier Inc. All rights reserved. Nuclear transfer is a technique used for producing cloned animals. As noted in Chapter 7, nuclear transfer consists of the removal of the nucleus from an oocyte and replacing it with the nucleus of a donor cell. Electric fusion and microinjection are commonly used for transferring nuclei into enucleated oocytes. After nuclear transfer, reconstructed embryos are artificially activated to initiate development and cultured in vitro until the optimal stage for embryo transfer. Animal cloning by nuclear transfer has been successful in several species, although the cloning efficiency is extremely low. Nuclear transfer technology has been useful for genetic modification of livestock because of several advantages conferred by the use of cultured cells compared with the pronuclear microinjection method. This chapter summarizes the protocols used for cloning in livestock.
