Precise evaluation of electrophoretic mobility distribution of nanoliposomes using microcapillary electrophoresis chips with sensitive fluorescent imaging

An electrophoretic mobility (EPM) measurement system that combines on-chip microcapillary electrophoresis with a low-noise EM-CCD camera was developed, and it enables the evaluation of the behavior of individual nanoparticles (∼100 nm) which have been difficult to measure accurately. This measurement system was applied for precise evaluation of the electro-phoretic behavior of nano-bioparticles (nanoliposomes), and EPM distribution of nanoliposomes was measured quantitatively. As a result, for the first time, the size-dependent broadening of the EPM distribution of nanoliposomes due to the compositional fluctuation of a small number of lipid molecules was revealed. Assuming random compositional fluctuation, the standard deviation of EPM of nanoliposomes was calculated, so that the experimental data and theoretical data are well fitted.
