Reconstitution of an epithelial-endothelial bilayer by the micromesh culture technique employing an artificial Matrigel basement membrane

This study reports a novel approach to reconstitute a bilayer cellular structure using a thin Matrigel basement membrane fabricated by the micromesh culture technique, an original technique developed by our group.We demonstrate the fabrication of a bilayer structure consisting of epithelial and endothelial cell layers interfaced with an artificial Matrigel basement membrane with a thickness of <1 μm. The approach is promising in the fabrication of second generation organ-on-a-chip systems, which permits direct cross layer cell-cell interaction for better mimicry of in vivo functionality.
