EVENTS ASSOCIATED WITH THE RAPID SEPARATION OF CELLS FROM DETACHED ROOTS OF AZOLLA-FILICULOIDES DEPEND ON PH

Detached roots of Azolla filiculoides more than 20 mm in length contained large cells, which had formed an abscission layer in the intact plant, at the proximal end. When the detached roots were placed in buffers at pH values above 6.5, the large cells expanded, rounded up and sprang away from the roots. The cell-separation process was complete within 3 min at pH 7.0. However, these events rarely occurred in buffers at a pH below 6.0.
The cell-separation process was not inhibited by cycloheximide or actinomycin D. However, pretreatment with papain markedly inhibited the process.
Some of the separated cells that had been generated by treatment at pH 7.2 were plasmolysed in response to high concentrations of mannitol. The concentrations of mannitol that plasmolysed the separated cells were much lower than those that plasmolysed the cells that remained attached to roots.
Electron microscopy revealed that the middle lamella area between the large cells had already been degraded to same extent when roots were collected. When the cells expanded and separated from the roots upon treatment with buffer at neutral pH, there was a final loss of the small amounts of remaining materials in the middle lamella area and swelling of the primary walls. The results suggest that treatment with buffers at neutral pH weakens both the adhesion between the large cells and the mechanical properties of the cell wall, with resultant expansion and subsequent separation of cells from roots. We propose that an enzyme(s), present in large cells and activated at neutral or weakly alkaline pH, participates in the modification of the walls.
