PTENは細胞増殖シグナルに対して強い活性化作用を示す脂質セカンドメッセンジャー、ホスファチジルイノシトール3,4,5-三リン酸(PIP3)を脱リン酸化する脂質ホスファターゼであり、PTENの不活性化はPIP3の蓄積を介して腫瘍病変の形成を引き起こす。悪性腫瘍の約半分はPTENの不活性化を伴っているとされているが、その分子機構にはまだわからない点も多い。本稿ではPTENの機能や制御機構に関するこれまでの知見を概説しながら、PTEN研究の歴史的背景や最新のトピックスを紹介する。(著者抄録)

PTENおよびPI3Kによって制御されるホスファチジルイノシトール3,4,5-三リン酸(PIP3)は、細胞増殖シグナルに対してきわめて強い活性化作用を有する脂質セカンドメッセンジャーであり、PTEN/PI3KによるPIP3制御系の異常は腫瘍病変形成を引き起こす重大な原因の1つである。本稿では、PTEN/PI3Kによる細胞内シグナル制御および腫瘍病変におけるその異常とともに、PTENのがん抑制因子としての機能、制御メカニズムに関して概説する。(著者抄録)
