To examine the fuunction of BMP4 on chondrogenesis in the mandibular process, we implanted hrBMP4-soaked beads and micro-injected Msx2 gene carrying Adenovirus (AdV-Msx2) into E1O mouse mandibular organ culture explants. BMP4-soaked beads induced ectopic cartilage at posterior portion of the explant but not at the anterior portion. BMP4 induced Msx2 and Sox9 expression simultaneously but the expression levels were very high at the anterior portion. Furthermore AdV-Msx2 micro-injection in the explant showed focal inhibition of Meckel's cartilage formation and Col2a1 expression. The findings suggest that BMP4 mediates expression of antagonistic functional transcriptional factors of Msx2 and Sox9 on the chondrogenesis.
