ポリグルタミン病は、CAGリピート配列の異常伸長という特徴的な遺伝子変異に起因する遺伝性疾患であるため、分子生物学を基盤とした病態研究が他疾患に先んじて進展してきた。遺伝子改変マウスの解析から、神経症状は神経細胞死に至る前の可逆性神経機能障害に起因することが明らかになった。その神経機能障害は、個々の神経細胞の自律的(cell-autonomous)な障害だけでなく、グリアなどを含む非細胞自律的(non-cell-autonomous)なネットワーク障害に起因すると考えられるようになった。そ...
